Breast cancer (BC) is the second common type of cancer in the world and has the leading mortality rate compared to other cancers affecting women. Leptin modulates multiple processes in the peripheral organs, acts as a mitogenic factor implicated in tumorigenesis suggesting to stimulate proliferation and survival. The aim of this study is to investigate the increase in Leptin and its possible role as BC biomarker. Expression of Leptin and Leptin receptor (LEPR) were evaluated, by qR-T PCR, in 65 female patients, 50 were diagnosed as breast malignant and 15 non-malignant, with no significant difference between the groups in age, BMI and waist/hip ratio. The blood samples were collected together with patients’ information and clinicopathological data, from KAU hospital, at the time of cancer diagnosis and prior to treatment. Total RNA was extracted from whole blood using PAXgene blood mRNA tubes and extraction kit. Serum Leptin level was also measured using ELISA. Our results show significant increase in Leptin mRNA (p=0.025) with no significant change in LEPR for malignant patients versus non-malignant. Furthermore, significant association were shown between the up-regulated Leptin expressions in malignant patients with their some clinicopathological parameters; tumor size (2-5cm), histotype (LCIN), negative vascular invasion, also hormone receptor phenotype such as: hormones receptor negative (ER-, PR-) and triple negative (ER-PR-HER2-). Leptin levels also showed significant correlation with age (p=0.04) and inverse correlation with BMI of non-obese (p= 0.01), as well as, with other patients characteristic parameters such as; premenopausal patients, no family history of BC or any other types of cancer and no physical activities. Consistent with observation by previous work in the group on increase Leptin expression in BC tissue, these results demonstrate increase serum and expression levels of Leptin in BC blood samples; furthermore, correlation with key molecular and pathological markers, may suggest a role for Leptin as a biomarker for BC. |
